The England captain reached the milestone on day two of the third Test against New Zealand at Trent Bridge, claiming his 250th Test wicket when Mitchell Santner gloved a short ball.
Kallis scored 13,289 runs and took 292 wickets in 166 Tests for South Africa.
Stokes, who is playing in his 122nd Test, has 7,228 runs to his name.
